Cytochalasin B
نویسندگان
چکیده
Cytochalasin B, a mold metabolite, has been shown to inhibit cytokinesis and motility (3), morphogenie development (18), endocytosis (5), and the transport of sugars (8-10, 12). I t has also been demonstrated that this drug inhibits the stimuatcd elaboration of extracellular substances from glands-the release of thyroxine from the thyroid gland after stimulation by thyroid-stimulating hormone (TSH) (19), and the release of o~-amylase from the parotid gland after stimulation by epinephrine (1). To date, the effects of cytochalasin B on other aspects of hormone-mediated responses have not been reported. Therefore, studies were initiated to study the effects of the drug on another hormonemodified system--the induction of tyrosine aminotransferase (E.C. 2.6.1.5) by insulin, cortisol, or N6-2CO-dibutyryl adenosine-31-5r-cyclic monophosphate. These studies were carried out with Reuber hepatoma (H-35) cells grown in culture.

P-75: The Effect of Hydrostatic Pressure in The Presence of Different Concentrations of Extracellular Calcium, Ethanol and Cytochalasin B on Parthenogenetic Activation of Mouse oocytes
Background: The parthenogenetic of oocytes is important in cloning research, as artificial activation of oocytes is an essential component of nucleus transfer protocols. Hydrostatic pressure can act as a mechanical stimulator that rearranges egg contents. Ethanol promotes a single intracellular Ca2+ increase of greater and longer amplitude than the initial increase observed at fertilization. In...
متن کاملOocyte Enucleation Phase in Mouse Cloning
Mouse oocytes were placed in Hepes-T6 containing 7.5 μg/ml cytochalasin B and 5μg/ml Hoechst prior to enucleation, and enucleation was performed on oocytes using the manipulator. The oocyte held in place by gentle suction of a holding pipette (oocyte diameter approx.70 μm). The metaphase spindle (arrow) is aspirated into an enucleation pipette (1).
متن کاملP-229: Chromosomal Analysis of Parthenogenetic Mouse Embryos Generated from In Vitro Activated Oocytes by Hydrostatic Pressure and Ethanol and Cytochalasin B
Background: Studies of preimplantation stage embryos by classic cytogenetic techniques have limitations, starting with the need for good metaphase stage when only one third of all analyzed embryos may show good quality metaphases. The incidence of chromosome anomalies in embryos produced by in vitro procedures is generally higher than that of embryos formed in vivo. Pressure specifically affect...
متن کاملDirect block of cloned hKv1.5 channel by cytochalasins, actin-disrupting agents.
The action of cytochalasins, actin-disrupting agents on human Kv1.5 channel (hKv1.5) stably expressed in Ltk(-) cells was investigated using the whole cell patch-clamp technique. Cytochalasin B inhibited hKv1.5 currents rapidly and reversibly at +60 mV in a concentration-dependent manner with an IC(50) of 4.2 microM. Cytochalasin A, which has a structure very similar to cytochalasin B, inhibite...
متن کاملDifferent aspects of cytochalasin B Blocked micronucleus cytome (CBMN cyt) assay as a comprehensive measurement tool for radiobiological studies, biological dosimetry and genome instability
It is now universally accepted that DNA is the main target for damages caused by physical and chemical genotoxicants. Although there are different methods to measure directly the induced DNA damages but due to fast repair processes in cellular environment, most of the damages would be repaired even before sampling, therefore processed DNA damages, i.e. damages left unrepaired after acting repai...
متن کاملCytochalasin inhibits the rate of elongation of actin filament fragments
Submicromolar concentrations of cytochalasin inhibit the rate of assembly of highly purified dictyostelium discoideum actin, using a cytochalasin concentration range in which the final extent of assembly is minimally affected. Cytochalasin D is a more effective inhibitor than cytochalasin B, which is in keeping with the effects that have been reported on cell motility and with binding to a clas...
